For those of you who have already taken the DAT, which preparation method worked for you? I'm thinking about getting the Kaplan Blue Book, but I'm not sure if it's necessary to read and practice the individual problems in every...single...chapter (that's a thick book!). Is this usually just how it has to be done, or is it a better idea to take a pre-test and just work on the areas I'm rusty in?

That's what I did. IMO, it' better than reading whole textbooks. :D

Obviously for something like the DAT, you want overkill. So here's what I'm doing. It may be a bit excessive, but I think it will get me the score I want (minimum of 19 on each section). Kaplan blue book, destroyer+odyssey, achiever, Crack DAT PAT and math, Campbell's Bio, Cliff's AP Bio, Schaum's Outline for Bio and Orgo, Barron's (only for PAT. Rest is useless), the KSF review (DAT+PAT Advanced +PAT extreme), Topscore Tests, oh and I have all the Kaplan online practice material. The point here is to go ape $hit and do so many problems that I go into the DAT feeling like nothing can surprise me.
