If you add 1 microliter from original sample of 5ml to 999 microliter. Than take 2 ul from that and add it to 990 microliter.

What is the dilution ratio?

1 ul added to 999 ul is a 1:1000 dilution. Taking 2 ul from that and adding it to 990 ul is a 2:992 dilution (or 1:496). We can multiply our dilution factors so our final result is a 1:496,000 dilution. In other words, for every 496,000 parts of a solution, one of those parts will be from your stock solution (the original 5ml). You could also say that your stock solution has been diluted 496,000 fold.
