I know that many medical schools require 2 semesters of English prior to matriculation. Do these courses actually have to come from the English department, or do Comparative Literature courses fulfill the requirement as well?
Also, what about AP credit for english courses?

Well a comparative lit course is an "English" course, so yes.

And any english or writing course will do, really.
I would not use your AP English credit to fulfill this requirement, take a class and learn something. Literature is fun!
