Generally, three letters of recommendation are required but check with each program to be sure. You can view specific requirements for individual programs by checking the residency directory at www.ashp.org
It is a good idea to include recommendations from at least two professors and possibly one from an employer so the program can evaluate you both as a student and as an employee.
I would add rotation preceptors to the mix. If you had a good clinical rotation, and you did well there, make sure you ask your preceptor for the letter. It is going to tell the program more about you in the clinical role than any professor who has only seen you in academic seeting would.
