My plan so far is to apply during the 2021-22 cycle, which would be 6 years from when I graduated undergrad in 2015. Is achieving a good MCAT score (515+) enough to indicate academic aptitude, or would it still be recommended to enroll in more upper division science classes? If so, would taking and doing well in just one upper div bio class the semester before applying, be sufficient? Thanks.

As long as you meet all the pre-requisite requirements, it is not necessary to take more classes. I graduated in 2009, and only took classes I needed to meet the requirements. Doing well on MCAT is a must.

Just check msar for each school you intend to apply to. Some (not many) explicitly state that prereqs must be take within x number of years of matriculation
