I go to a small school in Illinois and my school doesn't have a lab for principles of Organismal biology, they only have a lab for principles of biology. What should I do? Should I take the principles of biology lab twice? Is it necessary to have taken two labs for both biology courses?

most schools are very strict about taking two semesters of biology with lab, but very few demand that it must be general biology 101/102 (especially if your school doesn't have a lab for one). I would take the principles of biology course and ditch the organismal biology. Instead, just take any upper level course (with lab) of your choice to fulfill the second part of the biology requirement.

yeah you can usually replace an intro bio lab with an advanced bio lab, especially if you explain the circumstances.

I didn't take the lower level lab at my school (pre-research program had us skip it), and none of the schools cared that I'd used upper division labs to satisfy the requirements.
