Okay, i figured we could all bitch up a storm about that particular passage under one thread.. (i had form DM)

lemme start off by saying that the passage brought back memories of journal club days - i cant believe they'd put that level of depth in there - it usually takes 45 minutes just to orient yourself to the experimental goals ...

anyway, from what i can recall, the trp mutation caused the TRP protein not to bind to the PDZ, so no TRP band shows up on the trp lane.. so that second mutation had a band lacking in... PKC was it?
 
the western was crazy

proteins labelled on the left, mutates/control on the top. It is probably best to read it column by column, checking the two controls first.

Then you had to piece together how proteins bind together starting with the labelled antibody. If the sequence is Ab-X-Y then you would expect a mutate for X not to show X or Y but a mutate of Y to still show X.
 
The passage was insane as was the figure but I didn't think the questions were that bad. Only 2 or 3 of them dealt directly with the passage, the rest were background info.
