I took a semester off in Fall 2020 and did an internship, and ended up graduating in Fall 2023 instead of Spring 2023. I'm not quite sure how I would put my semester calculations, especially since I also took a few summer and winter classes. I wanted to ask if this assumption was correct: I would mark the semesters with the corresponding semester (ie. Fall 2021 would be my sophomore year 2nd semester, rather than junior year 1st semester) and put the academic years as they normally would have been (2021-2022 in the case of Fall 2021). Please let me know if that would be correct!

I do see the guide about interrupted coursework and how to count hours, but seeing as there is one semester off only, should I just proceed and mark it by semester and not credit hours like the handbook shows? I'm a little confused by the table given there.

I'm not sure I understand your question. AMCAS performs these calculations after you enter your grades and semester/terms properly.

When working on entering your coursework on AMCAS, first you will enter which colleges you attended and which terms you attended.
In your case you will enter Spring 2020 and the next term attended will be Spring 2021.
Enter your courses in the correct terms.
Don't worry about which term is "sophomore" or not, AMCAS will fix that up if needed during verification.

The AMCAS course listing section won’t ask you “why” there was a gap or one term not attended during college, but fairly often a med school will ask that in a secondary essay. Explain simply about the internship you did in that space
