1) does AMCAS verification basically mean that they only check that the grades you entered match your transcript?

2) Do they look at/contact ppl from your ECs at all (even a little bit), or is that part left up to med schools to do if they want?
1) Yes and to be sure you submitted all the transcripts from all post-high school, higher education institutions where you attempted coursework.

2) No, that's left up to individual schools to do, if they choose.
 
So what exactly happens if you submit your primary and forgot to include a course? Do they add it manually themselves when verifying your transcripts? Or is there some sort of delay in verification?

So long as you made less than 10 mistakes, they will enter the omissions. There would be no delay. If there are more than 10, they will send it back to you for correction, resulting in significant delays, as you will need to re-enter the verification queue on resubmission.

As an addendum should they make any changes you disagree with (even if it's gone ahead to your schools) you can submit an academic change request with some sort of proof. Usually quite accommodating, I got several course classifications changed back (to science) via this.
