Publication or significant number of grades. Since you had no previous research before, maybe you could update if you ever present research. I mean 100 hours is like, what? 2.5 weeks of working? Closer to the end of last cycle though, I just Hail Mary'd and sent update letters everywhere. Might work for you, but definitely did not get me off any wait list. That was updating with MD letter of recommendation and 4 months ish working as a scribe.
 
Since it's now late in the cycle, I'd say just start handing out those update letters like candy. We have nothing to lose 😀

Currently no A or IIs received. I continued to volunteer more and recently got a position in a clinical research lab and got about 100 hours. Obviously not a lot came from it but I’ve previously didn’t have any research experience so I’m wondering if it’s worth an update.
Nope, it's not, unless you're on a waiting list at a needy school school.

Good update material is a semester of great grades, winning a major award or scholarship, or getting (NOT submitting) a publication
