When I write letters for my students, I focus on the same things irrespective of how I know them: the general competencies medical schools have identified as important.

The lens I use to showcase those depends on whether I had the student in class, working in my lab, as an advisee, as part of a student organization I advise, etc. The strongest letters are for students I've had the most contact with in different circumstances.

So your PI should look through the guide for letter writers from AAMC, and identify the things they can talk about using specific examples from their interactions with you that speak to competencies important in medicine.

But it's very unlikely that your PI hasn't written letters before, so unless they are asking you for guideance, they probably already know.
