I just submitted a couple of update letters recently; basically, it was:
  1. A brief introduction to the letter, wishing a happy new year (in a non-casual way) and mentioning how I'm still extremely interested in X School of Medicine and consider it a top choice if I get an interview. For one of the schools, I changed it to my top choice because it is genuinely my #1... maybe you can write that for Mayo.
  2. My new grade from a class I took
  3. The new project I'm working on in my FT job I'm still working at since applying, and the new volunteering responsibilities I've been given.
  4. A thank you for considering my application and the opportunity to provide valuable updates that I feel are important to my candidacy to the X SoM, etc...
I hope this helps! I ran mine by three current med students who said it was a strong letter, but that feedback likely isn't as valuable as some of the AdComs on this forum. I hope they can give you some advice, too!

Thank you for the advice! Based on the way you structured your letter would you recommend placing the study abroad courses before the work update?
Yes, I would personally do that. It feels more like a "concrete" update and should probably go first. That's why I put my received grade before everything else.
 
In general, stay professional. While in some way, it feels like you're sending in postcards along your application journey, keep it to business when it comes to describing significant milestones you have passed.
