A letter from my PI, one science professor, one humanities prof... am I shooting myself in the foot?

Unfortunately, due to various reasons most of which were my fault, I didn’t get to know that many professors well enough. I’m a final semester senior and I’ll be applying this June. I asked my PI, a science professor, and a humanities professor for letters, but I haven’t been able to get letters from other science professors. I asked three others, one said no, one has since retired, and one pretty much ghosted me.

are these three letters sufficient?
 
You are in decent shape, assuming the letters are good.

PI should be a strong letter, especially if they know you well.
Science and non-science also check their respective boxes.

If you could supplement it with a strong volunteer/clinical letter (for example, mine was my EMS chief) then I think you could have all your bases covered.

This is entirely school dependent. Make sure you meet the LOR requirements of the schools you’re applying to.
 
I applied to 27 schools with a letter from my PI, a science professor, and a humanities professor and had no problems. But before applying, I did check each school’s letter requirements to make sure that I was in the clear. Except for a handful of schools, my letters fulfilled all the requirements.
 
This is exactly what I did. I was able to apply to most schools with no issue. I got an extra science letter for two schools, but I knew it was weak so I didn't send it where I didn't have to.
