So, I'm a 4th year DO student, I'll be taking USMLE Step 2 soon but forgot to note on my ERAS that I will be taking it. So, programs think I'm not taking it. Should I send out emails to let them all know that I will be? I received a rejection that was later rescinded and my app put on hold after I'd told them I was taking USMLE Step 2 (did not take Step 1). Thoughts?

There's no way to change that on your app, so the only way to update programs is to send them an email. I have no idea how many programs will care. If you want to let programs know, be polite, and short, explain when you're taking Step 2 and when you expect results back.
