does increase in intracellular calcium is enough to cause exocytosis of granules in cells e.g.like neuron ,leucocytes ?
or does the increase in intracellular calcium should necessarily come from the release from endoplasmic reticulum following stimulation through receptors?

let me put it this way,is increse in intracellular ca2+ enough to trigger exocytosis of vesicles( like in our neurons ) in blood cell like platelets?

Yes. ECF-->ICF can trigger exocytosis on its own. Or (as in cardiac myocytes) the influx of calcium is necessary to cause release of calcium from the SR.
