CD45RO, just like CD45RA, CD45RB... is an isoform of CD45 (LCA), which is a protein tyrosine phosphatase (remove phosphate).

another name for CD45RO is UCHL1, which is a deubiquitination enzyme (remove ubiquitin).

Does anyone know whether CD45RO is a phosphatase or a deubiqutination enzyme?
 
Tha latter. It is more formally known as ubiquitin carboxyl-terminal esterase.
 
in ALPS, the double negative T cells are also negative for CD45R0, in tht context it helps if you get a lymph node from a patient and you are suspecting ALPS
