cytokine question

Hey everyone
I'm going a little crazy trying to figure out the cytokines and i had a question...

FA says that IL-4 induces differentiation of Th2 cells and HY immuno says IL-2 induces their prolif (but nothing about IL-4)...any thoughts? Does this mean that IL-4 says "Be a Th2 cell" and IL-2 says "Multiply into lots of Th2 cells"?

Yes, that's exactly how I understand it. IL2 is a nonspecific mediator of T cell expansion, IL4 says be a TH2 helper and not a Th1 helper.

HY immuno also says something like "macrophage secretion of IL-12 acts synergistically with IL-2 to induce differentiation of Th1 and NK cells into Tc cells". Now am i missing something here? Can a Th1 cell or NK cell become a Tc cell?

Based on the antigen presented, different interleukins are produced by the Th cell. IL-4 induces differentiation of B cells, or activation rather, to secrete antibodies. IL-2 does two things. one, it activates Th-1 cells to multiply, AND it activates cytotoxic T cells
 
So far what you have heard is correct...IL-2 is a potent growth factor for primarily CD4 T cells, but CD8 T cells (aka Tc cells) do also proliferate nicely in the presence of IL-2. IL-4 is the prototype Th2-inducing cytokine--drives Th2 differentiation of naive CD4 T cells and helps with B cell differentiation.

As for IL-12, it is secreted by dendritic cells and macrophages, and helps induce 1) the differentiation of antigen-inexperienced, naive CD4 T cells into Th1 cells, 2) the activation of NK cells, and 3) the activation (in the appropriate context of MHC-antigen) of CD8 T cells. CD4 T cells nor NK cells never differentiate into Tc cells!
