While I don't think that letters of interest are worth much, it would not harm you to mention that your partner is in the same city as the school. If they care about your letter of interest (which they may not), it would strengthen your reasons for attending a school in that city.
 
You've not heard back as in not been offered an interview? That you have a loved one in the city in which the school is located will not matter at all. If you have a very strong application, have been interviewed, and are waiting for an admission decision, the fact that you are more likely than not to accept the offer if one is made might give you a little bump but then again, if you are a hot prospect in New York City, or Boston or Chicago, cities with a ton of med schools, you might still be a 50-50 or less in terms of accepting the offer depending on what other offers are on the table.

It is unlikely to move the needle, particularly if you are not married to your partner.
