Can you report graduate-level grades on AMCAS if you're not a degree seeking student? As a simple example, if I took a graduate listing for biochem and got an A, would my AMCAS graduate GPA be a 4.0 or non-existent? Has anyone gone this route to get another GPA on their application? (Obviously w/ more than just 1 credit)

Yes, any graduate-level class, regardless of whether or not it was taken towards a degree, will be counted in the AMCAS graduate GPA column. I would urge you to ask yourself why you want to take a graduate class or two.... I have two scenarios for you.

1) If your UG BCPM is strong (did well in all four pre-reqs and maybe a couple of upper-level classes), then yes, IMO a 4.0 in graduate-level biochem will be icing on your app.

2) If your UG BCPM is average/low, then taking a random graduate-level class and doing well will not be as beneficial to your app as say, taking an additional upper-level UG biochem and getting a 4.0 to boost your UG BCPM.

For the numerous reasons discussed in other graduate school threads, adcoms just are not as impressed with great grad school grades as they are with great UG grades. So, don't fool yourself into thinking that a 4.0 in a single graduate school class will wow the adcoms when they see your app. If your app is strong overall, it will be icing, if it isn't as strong, then don't expect it to help much. And yes, this is coming from someone who has a 30+ credit graduate degree with a very strong grad GPA.

My UG GPA is rubbish - but I'm going to peak out rather quickly because of the number of hours I have (did two degrees as an UG). So after I have over 200+ hours of undergraduate work, I was thinking taking a few grad classes would highlight my recent upswing in grades. Otherwise, another A in an UG class will only raise my GPA 2 hundreths of a point.

Grad classes wont do you any good unless you're applying to osteopathic schools. Unlike allopathic schools, osteopathic schools combine graduate and undergraduate grades.
