Only need 1 of the following:

1. Intro bio instructor

100-level course/about 200+ students. Wasn't considering him as I never went to office hours, and had pretty minimal contact with him. However, recently have ran into him several times, and he has surprisingly remembered my name (took course ~3 years back), has engaged me in very friendly/interested conversations, and appears to somehow know about or followed parts of my academic "career" (not sure if he's talked to another prof of mine, or what). Makes me think he would write a positive letter. Mostly, he's in the running due to extreme niceness and showing an interest, which are both pretty refreshing.

2. Genetics instructor

Upper-division genetics course, smaller than #1 (~80 people). Again, never went to office hours, but had some contact through after-class questions, a couple emails, etc. He seemed to like me well enough during class and, like #1, I have run into him since, and he has been complimentary and knows about a couple of small things I will not put on AMCAS (but would be nice to have on letter, though no guarantee he would).

(Does upper vs. lower-division course make a difference?)

3. Calc professor

Small class, so had plenty of 1-on-1, asked questions, only went to office hours a couple of times, and had conversations with him many times after class. Did well in class, and was one of a couple students he invited to join a more advanced section. Worried that some schools will not accept this for science, though.

Did very well in all 3 classes. If it makes any difference, #1 was online problem sets + multiple choice exams, #2 was extensive written problem-sets + essay/problem solving exams, #3 was....math.

Thanks for any input.

forget what subject it is. the adcoms just want your LOR that says this person is not afraid to ask questions, works hard, and has a very warm, outgoing personality. That's what the LOR is. It's a test to make sure you're not "Good Will Hunting" who only shows up to a few of the lectures, sleeps all the time in the library, but still aces all the tests. In other words, what kind of person are you?

That's my favorite movie!

I've had a related question for a while, which could help OP.

After accumulating a number of LOR's, can you speak to your pre-health advisor and ask, "Having seen the letters, would you be able to suggest which ones I should send based on the strength of their recommendations"?

Personally, I'm going to end up with (hopefully) four science professor LOR's. I'm going to need to choose which ones to use. If we can ask pre-health advisor which are the best written for us, OP should probably grab all three.

Why will your advisor be reading your letters? All of mine will be uploaded to AMCAS directly by the writer. Although, I would suspect that if your advisor does read them (applying to schools that don't use the AMCAS LWA?), informing you of their contents, or even how positive the letter is, would probably enter some grey area of confidentiality that they won't cross. Just a guess, though. Maybe it's worth asking. Won't work for me, unfortunately.

Bump. Anyone wanna pick a number?? Please?...

Get all three. Doesn't hurt to have an extra. Some schools, like Duke, require four letters.

If you pick one though, I would probably go with Genetics.
