Importance of LOR from a PI?

I plan on writing about the research that I have done as a significant experience but as of now I am not planning on asking my PI for a letter. He is very nice and liked me at the time, but it was a couple of years ago and I don't know how much weight his words will carry in the eyes of an AdCom today.

Will it seem suspicious if I write about research as a significant experience but do not have a letter associated with that time? I already have all of my academic/non-academic letters confirmed for this cycle. I have a total of 6 letters ready but I dont want to go beyond that per some other advice I have read on SDN.

Yes.
 
Might depend on the school, but for a research focused med school it would look suspicious if you have substantial research and no PI letter.
