I work in an ER and have one physician who is a DO I think would write a great letter of rec for me. Should I use it for my AMCAS application or should I get one from an MD and use his letter from my DO app only?

MD schools don't care about the type of medical degree. DO employment letters are fine.

DO would be perfectly fine unless they explicitly request an MD letter as a requirement (which none of the schools I applied to did).

Generally speaking, MD schools really don't want a "physician" letter. A few request a "clinical" letter that can come from anyone who supervised you. Some undergraduate schools seem to want one for a committee letter (a slick way to confirm shadowing).

DO schools love a DO letter, though.
