It's currently sept 25 and I only have 1 letter uploaded and sent to schools through ERAS. I've consistently run into my other letter writers in person who keep on telling me they will get in their letters. I have been keeping it cool, but is there a reason to freak out yet and really tell them firmly to get them in rush-fashion before October 1st? A more objective question: do programs NEED 3 letters before even considering to review my app and invite me to an interview or will they look at all my other stuff and read my letters pre/post interview to help make a decision in terms of ranking me?

If I was in your position, I would track your letter writers down and firmly but politely let them know you need your letters in by Oct 1st.

I know plenty of applicants getting invitations before Oct 1st without having all their letters in but I would think that most programs expect your application to be complete when Oct 1st comes around (with all letters in).
 
Varies by program. I can tell you I also only have 1 letter in and have 4 interviews. I will probably have my other letters in time though. Some programs will definitely filter incomplete applications.
