LOR question

Hi Guys,

I am first year student at UBC, Canada with a strong passion for Dentistry.

My first year undergrad GPA so far is 85.5% (we don't have on the 4 point scale).
I am thinking of applying to some schools next year in the US if my GPA is strong enough in second year.

I have read LOR requirements of some schools and they recommend getting 2 from the science faculty of the school.
-I can easily get one from a Organic Chem Prof because I am volunteering in his research lab over the summer and so he can easily give me letter of reference.

But I was wondering where did you guys the second LOR from? Did you guys actually work/volunteer for this second Prof or did you just take a course with him/her?
The reason that I am asking this question is because I am wondering if a Prof is going to give an excellent LOR if the only thing I have done with him or her is take a course.

Any help would be appreciated.
