one of my colleagues was told in private practice that the assistants under some work law must get a physical exam and blood work, is that really allowed and is it allowed for the practice to impose things like that on staff and the dentist?

Depends on a bunch of cirumstances, such as the state you're working in and the type of practice. If it's a "small" 1 doc, 1 assistant, 1 front desk and 1 hygienist, then unless the Doc is offering some medical insurabce plan where a physical and blood work is required for enrollment than no. If it's a larger practice or a chain, then there is a greater chance, once again depending on the type of plan being offered, that a physical and blood work might be required.

A physical and blood work just for employment (and not medical insurance and/or life insurance reasons), thats not very likely

its in NY but they arent offering them medical coverage which is weird.
