I e-submitted my application already and 2 0ut of 3 of my references are in. I don't think the person who still has to write the reference have time now so I asked someone else. Would it be possible for me to cancel that reference and add someone else even if I e-submitted already.

Yes. You can delete if they haven't replied after e-sbumitting, and then add a new one. The only ones locked in are completed references.
