Quick question regarding LoR

When the application cycle opens on june 2nd, how quickly can my professor send in the LoR? If I put her email in the system that day, would there be any issue with the turnaround time?

She is leaving for vacation on June 4th so I just wanted to sure that there won't be something I am overlooking.
