So I used AACOMAS service directly to get one of my profs to upload a LOR. However, today I decided I will use interfolio...do I have to resend that one LOR that is on AACOMAS now to interfolio? I am a bit confused how this works and what I should do/if I should do anything

You don't have to do anything. You can use both, unless they changed it since last year.
