This chart was given on a practice problem (sorry about the format - didn't let me copy it):
Fidelity of nucleotide incorporation on normal and mutant templates of ssDNA:
A = 1 (normal) | 100 (mutant)
C = 300,000 (normal) | 100,000 (mutant)
T = 3 (normal) | 50 (mutant)
G = .3 (normal) | 1 (normal)
Two sequences of ssDNA (one normal, and one mutated.. in which a guanine was oxidized to its oxidized form) were separately allowed to react with a DNA polymerase + free nucleotides, and after dsDNA formation, both versions were sequenced for DNA replication accuracy. The table above gave the results.
The purpose of the experiment was to see how the oxidized guanine template would affect replication accuracy. However, I don't think that pertains much to my question: how do you analyze the results?
I'm not sure what to make of them. For example, one question's answer was that both strands will have the same melting temperature... But I assumed they would be different since the rate of C-G incorporations are different. Seems my problem is just in the analysis of the results.
Could you put into simple(r) words what the table is trying to say?
