I know that TMDSAS has rolling applications. I was wondering, by what date should I apply (as in, by what date should I optimally apply)? What has everyone's advisers told them? Thanks!

You have until the 3rd week of June to consider yourself early. After mid-July, pray to God since people have already been notified about interviews and it will take 4 weeks for your app to be verified and actually cross their desk.

June 15th. And to tag on to the last poster, I began being notified the first week of july about interviews. You want to be in the first couple of batches. Good luck!

Is June 1st the first day we can submit the application? I took the May 11th MCAT and have been working really hard on the application so I can submit "early." Will submitting 6/1 vs. when I get my score make a big difference?

very soon. probably first week of june. Just beware that there are a lot of pepole that got screwed this past application cycle due to the pre-match acceptance system and so all the reapplicants are definitely gonna be pushing to get their applications in asap. good luck

Straight from an adcom at UTSW:

"For example, an application submitted now would not prompt a response until August, but one submitted in June or early July might also get a response in August, since that is when we are first able to consider applications. Thereafter, it might be as early as a few weeks after submission of the application. Nonetheless, we encourage everyone to apply early (i.e. in June or July)."

Again, I'd say around late June at the latest. If you have a better GPA and MCAT score, you have a bit of leeway (not too much). But of course, the most important thing you should do right now is to get transcripts in and have LORs done ASAP. Those 2 things take the longest to do and commonly hold up applications.
