I just finished taking Orgo lab post college because of the hassle of taking it while I was in school. I want to update my schools on the grade I received since I put it on the AMCAS application as a current/future course. What is the best way to update the school? Should I send a letter stating that I completed the course and received such and such grade? Send the official transcript to the admissions offices? Just wanted to know what worked well for anyone that has done this in the past or what people are planning to do. Thanks!

Send an e-mail with the update and submit an official transcript to every school that you applied to. Do be aware that many schools do not want any "updates" from you and will based their decision to invite for interview on you currently AMCAS. Since you listed current/in progress coursework, should you be accepted, they will want a final official transcript.

That sentence is very important, some school ONLY want you to send updated trascripts if you've completed substantial coursework after your AMCAS submission. So you don't want to annoy them with a single course. Definitely verify on a school by school basis.
