So does the 48hr drug screening need to be completed after you accept the offer or when the offer is received. I ask since a few people want I know want to wait a few days longer than 48hrs to hear back from other more desirable districts before accepting or declining.

Technically, tell your "friends" that the CRW offer is contingent on your "successful" test, so usually whenever you get the paperwork. This is state dependent on where the clock starts. For all of my jobs, the lab paperwork came with the hiring letter or in the case of the Feds, before even.
