That really is a broad question. I'd say definitely the ones in Kaplan book are essential, then if you're willing to spend more time, i'd go on and look at different resources to supplement your knowledge (topscore, more your old bio textbooks etc..)

i think it is important to memorize the classifications.
i have seen at least one or two questions on this subject on every practice DATs i have taken.
i think it is important to know the differences to do well.
