The application currently available on AMCAS is for the application cycle that began June 2009. You can set up an account and get into it, and even fill in the blanks, but it will reset in May. Then you'll have to re-enter everything again before applying in June 2010.
I don't believe they'll accept transcripts art this time, so hold off for awhile.
It's a fine idea to start writing your Personal Statement and (as mentioned) collecting Letters of Recommendation now.
You can't send in your transcripts until AMCAS opens up because you have to print out a sheet from AMCAS that has to accompany your transcripts when sent. AMCAS usually opens up in mid-May, and you can send your transcripts then.

Agree with above. Specifically you will be looking for AMCAS 2011, which is the application that will be used in June 2010 for the class entering in 2011.
