I'm familiar with "early decision" which is similar to what you are referring to. Basically, it's an application process that occurs BEFORE most application processes begin. Usually, if offered a spot, you have to commit to the school. Only select schools have this process though, so I would become familiar with each school's admissions process before you start applying.
Grade forgiveness basically replaces a bad grade with a new one in your GPA when you re-take the class. Seems like every school has a different policy on what they allow. Basically either search your school's website or ask an adviser. Both grades will still be on your transcript though. PharmCAS averages the grades. Some pharm schools only go for the highest grade.
You can take the PCAT in January, June, August, and October. When you take it is entirely up to you and when you feel prepared. If you take it early, you can always re-take it in time for admissions if you are unhappy with your score.

I should add that the October test is probably the last possible date you can take it for admission for the next year. Some schools do allow the January test though.

Just so I'm clear, if I'm hoping to apply for Fall 09, is taking the PCAT in June (or August) soon enough? Or am I stuck waiting until Fall '10?
I plan on taking the PCAT in June.
It would be fine to take the June or Aug 2008 PCAT if you want to apply for Fall 09. Most schools allow you to take it up to Oct 2008, and a few will even allow Jan 2009
