Yes, it is correct to say that ion-exchange chromatography separates components based on their charge, and how those charges interact with charges on the column. The answer to this question is still correct, but the language used to exclude answers B and C is incorrect. It should say something like "Answers B and C would separate proteins based on differences in charge, but the question stem specified the use of an affinity tag, so D is the better choice." Thanks for the heads up, I'll pass this along to someone who can update it.
