I don't have my histology notes on hand but I think basophilia is associated with lots of RER (high protein synthesis).

yeah i'd like to think so to but I've also read that alkaline phosphatase activity can be seen during " bone growth, healing fracture, acromegaly, osteogenic sarcoma, liver or bone metastases, leukemia, myelofibrosis, and rarely myeloma. Alkaline phosphatase is used as a tumor marker..."

Even if alkaline phosphatase activity was present during bone growth, the question asks what causes the BASOPHILIA. I think alkaline phosphatase activity is acidophilic.

The cytoplasm of osteoblasts appears to be basophilic via normal H&E stain due to presence of large amount of rough endoplasmic reticulum.

Osteoblasts do have alkaline phosphatase activity but abundant RER is responsible for basophilic staining.
