If you're thinking within the U.S., I'd def check out Alternative Spring Break if your school offers it. I did it for two years and loved it. My first year, I went down to North Carolina and volunteered for the Nature Conservancy (and got to stay in one of Rockefeller's old mansions!) The second year I stayed in state and we worked at a local domestic violence shelter. Both really interesting, eye-opening, and fun trips.
I also know a girl who did a health service trip in Nicaragua, through International Service Learning (ISL). She had a great experience--the website is here if you wanna check it out 🙂
I'm also going to promote International Service Learning. I went to Costa Rica on a medical service trip over spring break last year and LOVED it. Amazing experience. I would recommend ISL to any Pre-Med.
