I have looked into a lot of medical schools that ask for only one organic chemistry lab, however, some state they need two. For example UM Ann Arbor says they need two organic chemistry labs, if I only take one, will they not accept my application?

You need to complete all of the required courses before matriculation. If a school says they require it, they require it.
