Calling the Dean, yes or no?

I don't know if you can just call them up unless you have some sort of connection. I did have such a connection at one school and I actually met with a dean before applying at my dad's insistence. All I can say is that if you decide to do this, know what you're going to ask and make a postive impression. Don't go in there thinking that they will just talk and give you all the best advice without you asking questions.

Overall though, I wouldn't do it.
 
I'd write a letter. Not necessarily an "LOI" but something with additional questions. Maybe even just an e-mail.
 
It depends on the school. One of the schools I applied to actually encourage people that are waitlisted to make an appointment to talk to the dean. I did it, and although I don't think the meeting was very informative, at least he knows who I am now and that I have a real interest in the school. If your school doesn't encourage that kind of thing, send a letter or email. It won't hurt you, and even if it doesn't help, at least you feel like you are doing something.
 
For clarification purposes: are you on a waiting list or do you want to ask the dean some questions before even applying?

Ditto.

Like most things, it probably depends on the school. I took my application into the Dean of my state school because it is encouraged and he has no vote on the AdCom. He gave good advice and encouragement. I don't know of any waitlisters who have gone in, but many schools also encourage reapplicants to meet with the Dean to help correct mistakes and sell themselves better before reapplying.
