I would really appreciate it if someone could please clarify the following discrepancy:

in FA: linkage disequilibrium: measured in a population, not in a family (pg 88)
in HY CMB: linkage: can be measured only in family studies (pedigrees). Consequently, only family members can be used to determine linkage. (pg 57)

am i missing something?

thanks for your help

Probably because they're talking about two different things. Linkage is just how close two loci are to each other on a chromosome. The closer they are, the less likely they will be separated by a chiasmata in meiosis.

Linkage disequilibrium means that two alleles (not loci) are linked more/less often then they should be. So if mating isn't randomized, or if their is a mutation, or genetic drift then you will see some alleles appear more/less often than if it were a true random population.

FWIW I didn't see linkage or linkage mapping in FA or the new CMB 2nd ed., maybe not important anymore?
