I plan on having my DO LOR sent to my committee premed profile and let them send it to multiple schools. How would they make multiple copies of the LOR without opening the letter? Wouldn't unsealing the letter make it unofficial?

Letter services/committees can open, copy, and reseal letters at will. What they can't do is share the contents of the letter with the applicant if the applicant waves the right to read it (which is standard).

I used the INTERFOLIO service, and the DO was able to upload it electronically. I know there are letter services, and I'm sure many of them allow it to be uploaded electronically.

Even if not, siggy is absolutely correct.

Do you guys think it would be ok if I used both interfolio and my committee as my app organizers? My DO prefers to type my LOR so I'm going to use interfolio for only his LOR but the rest of my LORs will be from the committee profile. Would this be too confusing for schools to get things from both sources?

My friend and I were talking about this the other day. He suggested I ask the premed office if they can accept a copy of the letter from Interfolio. If so, you can have the actual letter sent to Interfolio and then foward it to the premed office.

That might get pretty confusing...try to either do it through interfolio or through your pre-med advising office, but not both.
