Could someone explain what's the mechanism of Conjugate vaccine please?

I think that in only polysaccharide vaccine, only IgM is made but not IgG.
Also I know that Conjugate vaccine would increase cell-mediated immunity.

However, I'm not clear about whether this cell-mediated immunity means it's done by stimulating CD4+ T cell or by only increasing Cytotoxic T cell activity..

many polysaccharides are not immunogenic so you for them to be a good vaccine you need to link them to something that causes a good immune response, generally a protein. The protein is called a hapten.

Cell mediated, at least as I understand it, refers to anything coming off Th1 cells while humoral to things coming off Th2 cells

Th1 --> macrophages (via IGN-y)
Th1 --> CD 8 cells (via IL-2)

Th2--> B cells (via IL-4, 5)

I couldn't have put it any more succinctly or better myself. I have a question about the HIb vaccine as I know that it uses some special sort of immunogenic capsule obtained from B. pertussis, or am I wrong? If so, wouldn't it initiate humoral immunity (via IL-4, IL-5, IL-6) and then release IL-10?
 
Yeah i am pretty sure it is an Ab to a specific capsule of H Flu (the B capsule) and uses a hapten in the form of pertussis, or diphtheria toxin or tetinoid toxins. The Abs bind H flu and prevent penetration of the most virulent strains (B strains). Note that kids can still get ear infections via the non-B capsule strains of H flu just not the meningitis.

So it should be humoral immunity

Word. I'm on top of Micro, I wish I could say the same about Gastro and Biochem 🙁
 
Just missed a UW question, so...

The protein from the HiB capsule is PRP (stands for something long). And of course usually paired with tetanus toxoid to increase immunogenicity.
