I am wondering--to what extent do we need to know about this? EK 1001 has barely any questions on in. I understand Young's double slit experiment, but do I need to know how to do problems involving the path difference?

They can show up on the MCAT, but I don't think that it's 'commit to memory' type equations. In reality though, when you understand wavelength/frequency and interference in depth, you'll be able to figure out the more basic path difference problems just by 'logicking' your way through the problem. If you get Young's double slit, you're probably good to go.
