Destroyer Note Cards?

Would making note cards of the DAT Destroyer's material be a waste of time? Or is going through the book over and over a more efficient way to study?

That all depends on how YOU learn best. For me, it easier to memorize something once I physically write it down. However, if you are a good learner just by reading and memorizing then you shouldn't have to write out flash cards.
For Destroyer, I keep a little notebook where I write out the entire answer to each question which, asides from drilling the information in my brain, it is easier to carry around.
