Do I start asking for LoR’s now, and other LoR questions

If my post-bacc goes as planned, the earliest I can apply is the 2022-2023 cycle, perhaps later. I’m currently in human anatomy, and I’m doing quite well and getting along with the lab instructor, so I was wondering if I should ask her for a LoR (won’t be able to ask my lecture prof since I never got a chance to really talk with him; his OH had conflicts).

But, that would mean one of my letters could potentially be 3+ years old. Is this ok? Or should I wait off until like 1-2 years before I apply?

Also, this is my first time asking for a LoR, ever.

1) Do I email the professor? Talk to her in person?

2) Does the professor have to have a PhD?

3) If she says yes, I ask her to send it to interfolio right?

I don’t know anything about this process, so my apologies for some dumb questions. Thank you.
 
Two suggestions..
When asking,I always suggest asking if: would they be willing, and could write a strong letter of recommendation.
Also, indicate that since it will be years from now, could they write it now and save it for sending in the future. I other words, if it is not someone with whom you’ll have ongoing contact as the basis of the letter, have them write it while you are still ‘fresh’ in their mind.
 
Unless you plan on working with them in the future, have them write it now and then update the date (and add any additional interval info) closer to when you apply. Letters can be stored in Interfolio. Always good to have backup letters regardless in case other letter writers fall through.
