Does a letter from research prof count as science prof?

I'm a bit concerned about using my letter from my research lab director as a science letter. I did research in a chemistry lab for a year and it's on my transcript as a research methods course but it's an satisfactory/unsatisfactory course. Also, I took the course for 0 credit so I didn't have to pay extra but it's still on my transcript.

My question: will this letter from my research director count as my 2nd letter from a science professor? It was written by my lab director who is an organic chem prof, but I didn't take him for orgo. I chose to get my letter from him rather than my bio prof because I felt that he knew me much better through research than my bio prof did through lecture.

I was just about ready to submit my application but then I noticed that certain schools like UMich specify that it must be a graded research course to count as a science letter - but mine was an S/U course so I'm not sure if that counts. Any help would really be appreciated, thanks.

I'm using a letter from my physics research instructor. But to be fair I have another science as a back up. I got an interview at mich last year with 2 bio and 1 physics research instructor. I think it counts, but call Michigan and ask. They're very phone friendly unlike other schools.
