does anyone know the purpose of Triton/acid urea gels? I am trying to find something that describes the theory behind it and how histones would run in such a gel, but I cannot find it anywhere. 🙁

I think the acid allows you to separate based on charge (protonating aa side groups), the urea allows you to separate based on size (denaturing the protein), and Triton allows you to separate based on hydrophobicity.

Don't know much about histones though.

H2A, H2B, and H3 variants present in human sperm simply
have similar mobilities in the acid-urea gel system although

thanks for your help
I have to write a page on triton/acid/urea, but I cannot find a place that explains it anywhere. I can find paper that use it though.
The power point slide really helps.

if i remember correctly, acetylation and phosphorylation cause a shift due to a change in charge, but methylation does not cause a change because it does not change the charge.
