If you already have an acceptance and get off a waitlist after the deadline for holding multiple seats, what is the procedure? How long can you hold on to the first acceptance seat, and the new spot from the waitlist?

actually...there is no rule set in stone. But you are expected to act as swiftly as possible. As well, I'm sure each school has its own policy.

at Emory, they told us that if you are accepted off the waitlist-you have 24 hours to decide.

Would it depend on when you got accepted from the waitlist?
If you get accepted of a wait list within a few weeks of the start of class, the person calling you often says they need to know within a few days. Understandable, as they want to fill their class roster ASAP.

Another interesting factoid, is that schools taking you off the waitlist after May 15, when you hold another acceptance, are supposed to advise your previous accepting school at the time of the offer (per MSAR).
