I have a 27 MCAT (10/10/7) and a 3.65 GPA. I'm thinking of just applying to all my state schools for this round. Then, if I don't get any acceptances, I'll re-apply next year. My question - will having a previous rejection significantly hurt my second application? I know that in a second year, I will be able to significantly beef up my application.

You will be guaranteed to be viewed as a re-applicant at the schools to which you have submitted a primary. In some states this is not a big deal.
That said, a strong first application is the best strategy.
